Has a team ever knocked off the defending National Champion in the NCAA Tournament and lose to what will be that year's champion in the same tournament? Will we be the first?

It happened last year. Arizona beat Duke, and then lost to UConn in their next game. Duke lost in the Sweet 16, though, not the first round.

2006--George Mason beats UNC, loses to Florida. 2004--Alabama beats 'Cuse, loses to UConn. No more digging; this appears to be a pretty common occurrence.
